Identificación de Cultivares y Líneas de Mejoramiento de Arroz de Chile Mediante Amplificación de Fragmentos Polimórficos (AFLP) Agricultura Técnica

Doce cultivares y líneas de arroz (Oryza sativa L.) desarrollados por el programa de fitomejoramiento del Instituto de Investigaciones Agropecuarias (INIA), Centro Regional de Investigación Quilamapu, fueron caracterizados genéticamente mediante amplificación de fragmentos polimórficos (AFLP). De 21 combinaciones ensayadas, sólo 16 fueron informativas, generando un total de 667 amplicones, 94 de ellos polimórficos (14,4%), con un rango entre 9 y 33% de polimorfismo por combinación. O objetivo deste trabalho foi construir um fenograma de trinta indivíduos de seis espécies de Spondias, amostradas em três diferentes locais, baseado em 120 marcadores AFLP das combinações de primers EcoR1/Mse1 para esclarecer as inter-relações entre as espécies, subsidiar a exploração das espécies como porta-enxerto e fornecer informações genômicas dessas espécies. O fenograma UPGMA do coeficiente de Jaccard apresentou boa definição, com valor co-fenético de 0,925 e localização externa de dois "outgroups" das espécies Mangifera indica e Schinopsis brasiliensis da família Anacardeaceae. Morphological and molecular characterization of common bean landraces cultivated in the semi-arid Mexican high plateau Boletín de la Sociedad

The objective of the current research was to characterize the common bean in agricultural fields planted with common bean landraces from the semi-arid Mexican high plateau using both morphological and amplified fragment length polymorphism (AFLPs) data. The morphological traits were discriminating and exhibited the clustering of 150 accessions based on the geographic origin and seed coat color. AFLP primer combinations exhibited a polymorphic range between 0.292 (E-AGG + M-ACT) and 0.375 (E-ACA + M-AGA). ABSTRACT Sugarcane orange rust caused by Puccinia kuehnii has recently become an important disease in sugarcane crops and its spread is causing great concern to growers. In this study, we analyzed spores from symptomatic orange rust sugarcane leaves collected in multiple locations in Cuba in a 4-year-period in order to characterize morphological traits of P. kuehnii, establish an adequate molecular technique to characterize it, and determine its infection court in sugarcane. It is commonly assumed that plants have more genetic diversity in their native range than in areas where they have been introduced due to founder effects. However, few studies have proven this assumption and included the comparison between non-native and native ranges. We analyzed AFLP fingerprint patterns of 149 individuals from five native (China) and five non-native (Argentina) populations of Cotoneaster franchetii, a shrub which successfully invades different habitats and forms extensive monospecific stands. Este trabalho teve por objetivo estudar a possível ocorrência de heterocariose por meio do pareamento de fungos provenientes de diferentes espécies de formigas cortadeiras coletados em diferentes regiões do Estado do Rio Grande do Sul, utilizando a técnica AFLP. Dez grupos de pareamentos foram realizados, sendo que em dois não ocorreu a fusão das colônias. Após ocorrerem os pareamentos, foram coletados discos de micélio da área de intersecção das colônias e das áreas extremas de pareamento, para a realização da extração do DNA e padronização. For the first time, the reduction in genetic diversity in the northernmost limit of natural occurence (southeastern Brazil) of Araucaria angustifolia in comparison with populations in the main area of the species continuous natural distribution (southern Brazil), was tested. O presente trabalho teve por objetivo verificar o efeito de diferentes períodos de luz ultravioleta sobre a similaridade genética do fungo simbionte de formigas cortadeiras pertencentes às espécies Acromyrmex ambiguus, Acromyrmex crassispinus e Acromyrmex lundi, através da técnica AFLP. Os fungos foram inoculados em meio de cultura Pagnocca e mantidos em câmaras climatizadas à temperatura de 25 + 1°C e escotofase de 24h, por um período de 15 dias. Potato cultivar identification using molecular markers PAB

The objective of this work was to evaluate a set of microsatellite markers for varietal identification and characterization of the most widespread potato cultivars in Brazil. The DNA from 14 potato cultivars was genotyped using microsatellite markers and the alleles were scored in silver-stained polyacrylamide gel. Twenty-four microsatellite markers were evaluated, and only one locus was monomorphic. Based on band patterns, a set of two microsatellites that were able to identify and differentiate all examined cultivars was obtained.

Somaclonal variation in plants regenerated by organogenesis from long-term cultured calluses of two diploid staminate genotypes of Asparagus officinalis cv. Argenteuil was characterized by plant phenotype, ploidy, meiotic behavior, pollen viability, fruit and seed set, and AFLP profiles. Phenotypic deviations from the donors were detected in foliage color, flower size, and cladode and flower morphology. Ploidy changes were observed in 37.8% of the 37 regenerants studied. Forty-four soybean genotypes with different photoperiod response were selected after screening of 1000 soybean accessions under artificial condition and were profiled using 40 SSR and 5 AFLP primer pairs. The average polymorphism information content (PIC) for SSR and AFLP marker systems was 0.507 and 0.120, respectively. Clustering of genotypes was done using UPGMA method for SSR and AFLP and correlation was 0.337 and 0.504, respectively. Cost reduction in plant breeding and conservation programs depends largely on correctly defining the minimal sample size required for the trustworthy assessment of intra- and inter-cultivar genetic variation. White clover, an important pasture legume, was chosen for studying this aspect. In clonal plants, such as the aforementioned, an appropriate sampling scheme eliminates the redundant analysis of identical genotypes. In the present study amplified fragment length polymorphism analyses (AFLP) was used to characterize 54 C. perfringens strains isolated from swine presenting diarrhea. Analysis of the results showed 29 distinct profiles with discriminatory index equal to 0.97. Partial correlation between the origin of the isolates and groups was drawn, and correlation was possible in only 18.5% of the samples.
